The city-boys came twice from behind to beat second tier side KGI on penalties, after a 2-all draw in regulation time.
The Bakau-based outfits were leading the game, but the city-boys put a spectacular performance in the closing stages to level the scores.
This pushed the game into a post-match penalty shootout which ended 3-2 in favour of Real de Banjul.
Real de Banjul will now clash with Brikama United in the quarter finals, and KGI are out and will hang their boots until next season.
The Bakau-based outfit will switch their attention to their remaining games, in order boost their hopes of staying in the second tier and avoid relegation to the third tier.
